Abstract
The study was designed to determine the expression of Follicle Stimulating Hormone Receptor (FSHR) in Kundhi buffalo bull Testicular tissue from post pubertal buffalo bull of about 2 years of age was collected from local slaughter house of Tandojam, Sindh Pakistan. After separation from scrotum, the testes were checked for their normal status. After confirmation of healthy testes, the samples were collected and transferred under ice to laboratory. After making small pieces, 30 mg of testicular tissue was taken for extraction of DNA. The commercial DNA extraction kit (Fermentas, Europe) was used for yielding the DNA; immediately after collection, the extracted DNA was stored at -20ºC The measurement of concentration (in ng/µl) and its absorbance ratio (260/280 nm; purity) was checked by Nano Drop 1000. For expression of follicle stimulating hormone receptor primers were designed from available sequence published online at pubmed. The corresponding DNA was used for amplification in real time PCR for expression of FSHR using SYBR Green method.
The follicle stimulating hormone receptors are known to be expressed in post pubertal buffalo bull when corresponding DNA was amplified in real time thermal cycler.
